The device requires one long microchannel (with at least one inlet and one outlet) and a temperature control element with at least two different temperature regions: one for denaturation and one for primer annealing/extension. The reaction flows though these regions in an alternating fashion to achieve the number of cycles required for the PCR reaction. (The reaction can be performed in droplets to avoid problems from a parabolic flow profile in the microchannel.) The main advantage of PCR on chip is a much faster heat transfer (resulting from the large surface to volume ratio) and therefore a much shorter overall reaction time. (Additional advantages from the use of droplets are a better reliability and the possibility to amplify a different DNA sequence in each droplet)
